Quantification of NADPH balance during adipogenesis

چکیده
Background NADPH provides reducing power for macromolecule synthesis and antioxidant defense. The pathways used to make NADPH under different physiological circumstances remain unclear. In growing cells, much NADPH production is coupled to nucleotide synthesis via the oxidative pentose phosphate and folate pathways. Here we examine, using isotope tracers and flux analysis, NADPH production routes in adipocytes.

Acetylation of malate dehydrogenase 1 promotes adipogenic differentiation via activating its enzymatic activity.
Acetylation is one of the most crucial post-translational modifications that affect protein function. Protein lysine acetylation is catalyzed by acetyltransferases, and acetyl-CoA functions as the source of the acetyl group. Additionally, acetyl-CoA plays critical roles in maintaining the balance between carbohydrate metabolism and fatty acid synthesis. Here, we sought to determine whether lysi...
متن کاملFifty Percent Ethanolic Extract of Momordica charantia Inhibits Adipogenesis and Promotes Adipolysis in 3T3-L1 Pre-Adipocyte Cells
Background: Natural products have gained importance recently for the treatment of obesity and its complications, partly because of the side effects of modern drugs.Hence, we aimed to study and compare the effect of varying concentrations of Momordicacharantiaon adipogenesis and adipolysis using 3T3-L1 pre-adipocyte cell lines. Methods: 3T3-L1 pre-adipocytes were procured from the National Ce...
متن کاملUric Acid-Induced Adipocyte Dysfunction Is Attenuated by HO-1 Upregulation: Potential Role of Antioxidant Therapy to Target Obesity
Increased uric acid levels have been implicated in the pathogenesis of metabolic syndrome. To examine the mechanisms by which this occurs, we hypothesized that an increase in heme oxygenase 1, a potent antioxidant gene, will decrease uric acid levels and adipocyte dysfunction via suppression of ROS and xanthine oxidase (XO) levels. We examined the effect of uric acid on adipogenesis in human me...
متن کاملGhrelin in growth and development.
Exogenous administration of ghrelin increases caloric intake and stimulates growth hormone (GH) secretion, two effects that are mediated through binding of ghrelin to the GH secretagogue receptor (GHS-R). In addition, ghrelin is thought to inhibit adipogenesis by GHS-R-independent mechanisms. In adults, ghrelin is mainly produced by the stomach. In contrast, in the fetal and early postnatal per...
متن کاملAdaptive changes of redox status in rat brain tissues due to decimeter microwave irradiation
Electromagnetic waves affect living organisms and it is of great interest for wide interaction of new sources with a diversity of frequencies and powers to life of people. In the last few years, many authors have proposed that the biological effect of electromagnetic fields in both the high-frequency and low-frequency ranges are connected with oxidative processes in tissues. Studying the change...
